Le signal de fluorescence augmente de fa\u00e7on directement proportionnelle \u00e0 la quantit\u00e9 de produit PCR. Le syst\u00e8me LightCycler\u00ae 480 Instrument II est disponible dans notre laboratoire.<\/p>\n<\/li>\n<li>\n<p class=\"font_8\">La <b>MLPA <\/b>est une technique PCR multiplex facile \u00e0 utiliser pour d\u00e9tecter les modifications du nombre de copies dans plus de 50 r\u00e9gions d&rsquo;une s\u00e9quence d&rsquo;ADN ou d&rsquo;ARN g\u00e9nomique qui peut distinguer m\u00eame une seule modification de nucl\u00e9otide.<\/p>\n<\/li>\n<li>\n<p class=\"font_8\"><b>S\u00e9quen\u00e7age de Sanger <\/b>D\u00e9couverte en 1977 et longtemps consid\u00e9r\u00e9e comme l&rsquo;\u00e9talon-or, cette m\u00e9thode est bas\u00e9e sur la terminaison classique des cha\u00eenes \u00e0 l&rsquo;aide de ddNTP. L&rsquo;appareil de s\u00e9quen\u00e7age ABI 3500 avec 8 capillaires est utilis\u00e9 dans notre laboratoire.<\/p>\n<p>  a) PCR primaire<\/p>\n<p>  b) R\u00e9action de s\u00e9quen\u00e7age<\/p>\n<p>  c) \u00c9lectrophor\u00e8se capillaire et \u00e9valuation informatis\u00e9e<\/p>\n<\/li>\n<li>\n<p class=\"font_8\"><b>S\u00e9quen\u00e7age de nouvelle g\u00e9n\u00e9ration (NGS) : <\/b>Le NGS est une m\u00e9thode de s\u00e9quen\u00e7age en profondeur bas\u00e9e sur le s\u00e9quen\u00e7age simultan\u00e9 de millions de fragments d&rsquo;ADN ou d&rsquo;ARN provenant d&rsquo;un \u00e9chantillon. L&rsquo;objectif est d&rsquo;\u00e9valuer le nombre et l&rsquo;organisation des chromosomes et la pr\u00e9sence\/absence de s\u00e9quences d&rsquo;ADN.<\/p>\n<p class=\"font_8\">Dans cette section, des techniques telles que les techniques FISH et Array-CGH sont utilis\u00e9es ;<\/p>\n<ol class=\"font_8\">\n<li>\n<p class=\"font_8\">La <b>FISH <\/b>est une m\u00e9thode qui utilise des \u00ab\u00a0sondes\u00a0\u00bb con\u00e7ues pour se lier \u00e0 certaines r\u00e9gions des chromosomes et \u00e9mettre des radiations lorsqu&rsquo;elles sont li\u00e9es. Elle comprend les \u00e9tapes de la liaison \u00e0 la r\u00e9gion cible et de l&rsquo;analyse du rayonnement produit par la liaison \u00e0 travers des filtres appropri\u00e9s dans le microscope. L&rsquo;aneuplo\u00efdie chez les individus et les embryons est reconnue par l&rsquo;analyse FISH dans de nombreux \u00e9chantillons tels que le sang, le liquide amniotique, les embryons et le mat\u00e9riel d&rsquo;avortement, et cette m\u00e9thode est \u00e9galement appliqu\u00e9e au cancer et aux maladies h\u00e9matologiques.<\/p>\n<\/li>\n<li>\n<p class=\"font_8\"><b>Array-CGH (caryotypage mol\u00e9culaire)<\/b> L&rsquo;A-CGH est une technique, \u00e9galement connue sous le nom de caryotypage mol\u00e9culaire, qui permet de d\u00e9tecter des irr\u00e9gularit\u00e9s g\u00e9nomiques au niveau de l&rsquo;ADN et sur l&rsquo;ensemble du g\u00e9nome. Cette m\u00e9thode pr\u00e9sente l&rsquo;avantage de pouvoir d\u00e9tecter des modifications du nombre de copies comprises entre 50 et 100 Kb, de ne pas n\u00e9cessiter de plaque de m\u00e9taphase et de ne n\u00e9cessiter qu&rsquo;une petite quantit\u00e9 d&rsquo;\u00e9chantillon d&rsquo;ADN pour l&rsquo;\u00e9tude. La plateforme Agilent Microarray est disponible dans notre laboratoire.<\/p>\n<\/li>\n<\/ol>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/section>\n\t\t\t\t<div
