Bei den hier durchgef\u00fchrten Analysen werden die zu untersuchenden Chromosomen aus teilbaren Zellen wie Blut, Haut, Amnionzellen und einigen Geweben wie Knochenmark, Chorionzotten und Hoden gewonnen.<\/p>\n<p class=\"font_8\">Die konventionelle Zytogenetik gilt nach wie vor als \u201eGoldstandard\u201c in der pr\u00e4natalen und postnatalen Diagnostik, da sie Informationen \u00fcber alle mikroskopisch nachweisbaren Anomalien des gesamten Genoms liefert.<\/p>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/section>\n\t\t\t\t<section class=\"elementor-section elementor-inner-section elementor-element elementor-element-9857882 elementor-section-boxed elementor-section-height-default elementor-section-height-default\" data-id=\"9857882\" data-element_type=\"section\">\n\t\t\t\t\t\t<div class=\"elementor-container elementor-column-gap-default\">\n\t\t\t\t\t<div class=\"elementor-column elementor-col-100 elementor-inner-column elementor-element elementor-element-11bc552\" data-id=\"11bc552\" data-element_type=\"column\">\n\t\t\t<div class=\"elementor-widget-wrap elementor-element-populated\">\n\t\t\t\t\t\t<div class=\"elementor-element elementor-element-0381391 elementor-widget elementor-widget-heading\" data-id=\"0381391\" data-element_type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Molekulargenetisches Labor<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-b7e028e elementor-widget elementor-widget-text-editor\" data-id=\"b7e028e\" data-element_type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t\t\t\t\t\t<p>Untersucht werden die Bestimmung von Mutationen und\/oder Genotypen, die zu einer Erbkrankheit f\u00fchren, die Tr\u00e4geranalyse einer genetischen Krankheit und die Analyse der Anf\u00e4lligkeit f\u00fcr multifaktorielle Krankheiten und Einzelgenkrankheiten (wie Thalass\u00e4mie, FMF).<\/p>\n<p class=\"font_8\">In diesem Abschnitt verwendete Techniken;<\/p>\n<ol class=\"font_8\">\n<li>\n<p class=\"font_8\"><b>Real-Time PCR<\/b> ist eine Technik zum Nachweis und zur Quantifizierung von mutierter DNA in Echtzeit unter Verwendung von Fluoreszenzfarbstoffen. Das Fluoreszenzsignal steigt in direktem Verh\u00e4ltnis zur Menge des PCR-Produkts. Das LightCycler\u00ae 480 Instrument II System ist in unserem Labor verf\u00fcgbar.<\/p>\n<\/li>\n<li>\n<p class=\"font_8\"><b>MLPA <\/b>ist eine einfach zu handhabende Multiplex-PCR-Technik zum Nachweis von Kopienzahlver\u00e4nderungen in mehr als 50 Regionen einer genomischen DNA- oder RNA-Sequenz, bei der sogar eine einzelne Nukleotidver\u00e4nderung unterschieden werden kann.<\/p>\n<\/li>\n<li>\n<p class=\"font_8\"><b>Sanger-Sequenzierung <\/b>Diese 1977 entdeckte Methode, die lange Zeit als Goldstandard galt, basiert auf dem klassischen Kettenabbruch mit ddNTP. In unserem Labor wird ein ABI 3500 Sequenzierger\u00e4t mit 8 Kapillaren verwendet.<\/p>\n<p>  a) Prim\u00e4re PCR<\/p>\n<p>  b) Sequenzierungsreaktion<\/p>\n<p>  c) Kapillarelektrophorese und computergest\u00fctzte Auswertung<\/p>\n<\/li>\n<li>\n<p class=\"font_8\"><b>Sequenzierung der n\u00e4chsten Generation (Next Generation Sequencing, NGS): <\/b>NGS ist eine Sequenzierungsmethode mit gro\u00dfer Tiefe, die auf der gleichzeitigen Sequenzierung von Millionen von DNA- oder RNA-Fragmenten aus einer Probe beruht. Ion Genestudio S5 Sequenziersystem, Illumina MiSeq und NextSeq Plattformen sind in unserem Labor verf\u00fcgbar.<\/p>\n<\/li>\n<\/ol>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/section>\n\t\t\t\t<section class=\"elementor-section elementor-inner-section elementor-element elementor-element-8a98169 elementor-section-boxed elementor-section-height-default elementor-section-height-default\" data-id=\"8a98169\" data-element_type=\"section\">\n\t\t\t\t\t\t<div class=\"elementor-container elementor-column-gap-default\">\n\t\t\t\t\t<div class=\"elementor-column elementor-col-100 elementor-inner-column elementor-element elementor-element-81c75af\" data-id=\"81c75af\" data-element_type=\"column\">\n\t\t\t<div class=\"elementor-widget-wrap elementor-element-populated\">\n\t\t\t\t\t\t<div class=\"elementor-element elementor-element-6d0536c elementor-widget elementor-widget-heading\" data-id=\"6d0536c\" data-element_type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Labor f\u00fcr molekulare Zytogenetik<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-8e7c4ec elementor-widget elementor-widget-text-editor\" data-id=\"8e7c4ec\" data-element_type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t\t\t\t\t\t<p class=\"font_8\">Zytogenetische und molekulare Methoden werden gemeinsam eingesetzt. Ziel ist es, die Anzahl und Organisation der Chromosomen sowie das Vorhandensein bzw. Fehlen von DNA-Sequenzen zu beurteilen.<\/p>\n<p class=\"font_8\">In diesem Abschnitt werden Techniken wie FISH- und Array-CGH-Techniken eingesetzt;<\/p>\n<ol class=\"font_8\">\n<li>\n<p class=\"font_8\"><b>FISH <\/b>ist eine Methode, bei der \u201eSonden\u201c verwendet werden, die sich an bestimmte Regionen der Chromosomen binden und bei Bindung Strahlung aussenden. Es umfasst die Schritte der Bindung an die Zielregion und der Analyse der von der Bindung ausgehenden Strahlung durch geeignete Filter im Mikroskop. Aneuploidie bei Individuen und Embryonen wird durch die FISH-Analyse in vielen Proben wie Blut, Fruchtwasser, Embryonen und Abortmaterial erkannt, und diese Methode wird auch bei Krebs und h\u00e4matologischen Erkrankungen angewendet.<\/p>\n<\/li>\n<li>\n<p class=\"font_8\"><b>Array-CGH (Molekulare Karyotypisierung)<\/b> A-CGH ist eine Technik, die auch als molekulare Karyotypisierung bezeichnet wird und mit der genomische Unregelm\u00e4\u00dfigkeiten auf DNA-Ebene und im gesamten Genom nachgewiesen werden k\u00f6nnen. Diese Methode hat den Vorteil, dass sie Ver\u00e4nderungen der Kopienzahl im Bereich von 50-100 Kb nachweisen kann, keine Metaphasenplatte ben\u00f6tigt und eine geringe Menge an DNA-Proben f\u00fcr die Untersuchung ausreicht. Die Agilent Microarray Plattform ist in unserem Labor verf\u00fcgbar.<\/p>\n<\/li>\n<\/ol>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/section>\n\t\t\t\t<div class=\"elementor-element elementor-element-7cafb9a elementor-widget elementor-widget-image\" data-id=\"7cafb9a\" data-element_type=\"widget\"
